ABSTRACT

Abstract The impact of fish oil concentration on the oxidative stability of microcapsules through the spray drying process using chitosan and maltodextrin as wall material was studied. Emulsions were prepared with different Tuna fish oil (TFO) content (TFO-10%, TFO20%, TF030% TF0-40%) while wall material concentration was kept constant. Microencapsulated powder resulting from emulsion prepared with high fish oil load have high moisture content, wettability, total oil and low encapsulation efficiency, hygroscopicity and bulk tapped density. Oxidative stability was evaluated periodically by placing microcapsules at room temperature. Microcapsules prepared with TFO-10% presented high oxidative stability in terms of peroxide value (2.94±0.04) and anisidine value (1.54±0.02) after 30 days of storage. It was concluded that optimal amounts of fish oil for microencapsulation are 10% and 20% using chitosan and maltodextrin that extended its shelf life during study period.


Resumo Foi estudado o impacto da concentração de óleo de peixe na estabilidade oxidativa de microcápsulas por meio do processo de secagem por atomização, utilizando quitosana e maltodextrina como material de parede. As emulsões foram preparadas com diferentes teores de óleo de atum (TFO) (TFO-10%, TFO20%, TF030% TF0-40%), enquanto a concentração de material de parede foi mantida constante. O pó microencapsulado resultante da emulsão preparada com alta carga de óleo de peixe tem alto teor de umidade, molhabilidade e óleo total e baixa eficiência de encapsulação, higroscopicidade e densidade extraída a granel. A estabilidade oxidativa foi avaliada periodicamente colocando microcápsulas à temperatura ambiente. As microcápsulas preparadas com TFO-10% apresentaram alta estabilidade oxidativa em termos de valor de peróxido (2,94 ± 0,04) e valor de anisidina (1,54 ± 0,02) após 30 dias de armazenamento. Concluiu-se que as quantidades ideais de óleo de peixe para microencapsulação são de 10% e 20% usando quitosana e maltodextrina que prolongaram sua vida útil durante o período de estudo.

ABSTRACT

Abstract Aim: To investigate if treadmill exercise (Ex) associated with fish oil (FO) supplementation during lactation would influence the biochemical profile as well as the oxidative balance in the hearts of male juvenile rats. Methods: Fifteen days-old rats were submitted to a daily moderate Ex training (based on their maximal running capacity) and FO supplementation for 4 weeks. Forty-eight hours after the last exercise session, blood fasting glucose and lipid profile were assessed according to the manufacturer's recommendations, while the oxidative status of the hearts was evaluated via colorimetric and absorbance-based assays. Results: FO associated with Ex decreased triglycerides (TG-79.27 ± 5.75 to 60.24 ± 6.25 mg/dL) and very-low-density lipoprotein cholesterol levels (VLDL-15.85 ± 1.15 to 12.05 ± 1.25 mg/dL) when compared to sedentary animals. FO, alone, reduced atherogenic index (AI- 1.14 ± 0.03 vs. 1.01 ± 0.04 a.u) while increased high-density lipoprotein cholesterol (HDL-43.90 ± 2.50 vs. 59.43 ± 3.15 mg/dL) of sedentary animals. Additionally, both Ex (67.3 ± 13.5 nmol/mg prot) and FO supplementation (56.6 ± 5.5 nmol/mg prot) decreased the oxidative damage to lipids in non-trained animals (105.8 ± 10.8 nmol/mg prot). The interventions also protected the protein content from oxidative stress (Ex- 5.15 ± 0.46; FO- 4.5 ± 0.5; and vehicle sedentary-7.3 ± 0.6 µmol/mg prot), while increasing the antioxidant defense and oxidative metabolism. Conclusion: Our findings suggest that intervention in juvenile rats can improve cardiac metabolism. These are the first findings to show the positive effects of the association between FO and moderate treadmill Ex during the critical period of development. We believe these results can drive early-life origins of heart disease through different avenues and, possibly, assist the development of a heart disease prevention program as well as an adjunctive therapeutic resource.

ABSTRACT

SUMMARY: Rodents are animals extensively used in biomedical and nutrition research, a necessary step before the research in humans. The composition and type of administration of the experimental diets are relevant and should be thought, considering each type of animal used in the research. It is particularly important to consider, among others, the metabolic differences between species and food needs in macro- and micronutrients to avoid possible bias. The American Institute of Nutrition (AIN) made recommendations for rodents, adapted to the period of growth (AIN-93G), which are pivotal in fetal programming studies. The experiments can be compared among different studies and better translated into humans, considering these limitations in the nutrition of parents and offspring. The review addresses different compositions of experimental food for rodents during development with the ability to induce fetal programming in the offspring and chronic diseases in adulthood due to the nutrition of the mother and father. The 'developmental origins of health and disease' (DOHaD) concept due to maternal nutrition is commented considering the protein restriction, vitamin D restriction, obesity, and intake of fructose or fish-oil. The 'paternal origins of health and disease transmission' (POHaD), because of the nutritional state of the father, were also analyzed in the review, primarily considering the obesity of the father. The review proposes some diet compositions to experimental research considering varied nutritional situations, hoping to assist young researchers or researches not familiar with experimental diet manipulations in the elaboration of the projects.


RESUMEN: Los roedores son animales utilizados frecuentemente en la investigación biomédica y nutricional, un paso necesario antes de la investigación en humanos. La composición y el tipo de administración de las dietas experimentales son relevantes y se debe considerar cada tipo de animal utilizado en los estudios. Es particularmente importante considerar las diferencias metabólicas entre las especies y las necesidades alimentarias de macro y micronutrientes para evitar posibles sesgos. El Instituto Americano de Nutrición (AIN) estableció recomendaciones para los roedores, adaptadas al período de crecimiento (AIN-93G), que son fundamentales en los estudios de programación fetal. Los experimentos se pueden comparar entre diferentes estudios y aplicar en humanos, considerando estas limitaciones en la nutrición de padres e hijos. La revisión aborda diferentes composiciones de alimentos para estudios experimentales en roedores durante su desarrollo, con la capacidad de inducir programación fetal en la descendencia y enfermedades crónicas en la adultez, considerando la nutrición de los padres. El concepto de 'orígenes del desarrollo de la salud y la enfermedad' (DOHaD) debido a la nutrición materna se comenta considerando la restricción de proteínas, la restricción de vitamina D, la obesidad y la ingesta de fructosa o aceite de pescado. Los 'orígenes paternos de la salud y transmisión de enfermedades' (POHaD), debido al estado nutricional del padre, también fueron analizados considerando principalmente la obesidad del padre. La revisión propone algunas composiciones dietéticas a la investigación experimental considerando situaciones nutricionales variadas, con la esperanza de ayudar a jóvenes investigadores o investigadores no familiarizados con las manipulaciones experimentales de la dieta en la elaboración de los proyectos.

Objetivos: o óleo de peixe, rico em ácido eicosapentaenoico (EPA), mostrou benefícios renoprotetores em modelos animais de doenças crônicas. O objetivo deste trabalho foi avaliar os efeitos da suplementação com óleo de peixe três vezes mais concentrado em EPA (EPA 3X) na função renal de ratos e a eficiência desta suplementação por um curto período de tempo. Metodologia: ratos Wistar (n=16) foram divididos em dois grupos, controle (C) e suplementado (CO) por 14 dias com óleo de peixe (EPA 3X). Foram determinados os seguintes parâmetros: ganho de peso, ingesta de água e ração, fluxo urinário, proteinemia, trigliceridemia, osmolaridade, clearance de creatinina e fração de excreção de sódio. Resultados: os ratos suplementados tiveram níveis de triglicerídeos no plasma 58,4% menores que os ratos controle (p=0,0239), entretanto os restantes dos parâmetros sistêmicos avaliados não apresentaram alterações. A suplementação com óleo de peixe não alterou o fluxo urinário. Entretanto, no grupo CO identificou-se um aumento na filtração glomerular, com um incremento em média de 67,8% no clearance de creatinina em relação ao grupo C (p=0,054). A excreção média de proteínas totais na urina e a fração de excreção de sódio foram semelhantes entre os grupos. Conclusão: a suplementação com óleo de peixe três vezes mais concentrado em EPA por um período curto de tempo (quinze dias) provocou alterações significativas tanto em parâmetros sistêmicos (redução significativa na trigliceridemia) quanto na função renal de ratos controle (incremento na filtração glomerular) indicando a eficiência desta suplementação.


Objective: Fish oil, rich in eicosapentaenoic acid (EPA), has shown renoprotective benefits in animal models of chronic diseases. The objective of this work was to evaluate the effects of supplementation with fish oil three times more concentrated in EPA (EPA 3X) on renal function of rats and the efficiency of this supplementation for a short period of time. Methodology: Wistar rats (n=16) were divided into two groups, control (C) and supplemented (CO), supplemented for 14 days with fish oil (EPA 3X). The following parameters were determined: weight gain, water and feed intake, urinary flow, proteinemia, triglyceridemia, osmolarity, creatinine clearance and fractional sodium excretion. Results: The supplemented rats had plasma triglyceride levels 58.4% lower than control rats (p = 0.0239), however the rest of the evaluated systemic parameters did not change. Fish oil supplementation did not alter urinary flow. However, in the CO group an increase in glomerular filtration was identified, with an increase of 67.8% on average in creatinine clearance compared to group C (p = 0.054. The mean excretion of total proteins in the urine and the fractional sodium excretion were similar between groups. Conclusion: Supplementation with fish oil three times more concentrated in EPA for a short period of time (fifteen days) caused significant changes both in systemic parameters (significant reduction in triglyceridemia) and in the renal function of control rats (increase in glomerular filtration) indicating the efficiency of supplementation.

ABSTRACT

Perception of asthma can affect the patient's self-management and outcome treatment. The outcome is not onlydetermined by symptoms, but also by illness perception. Omega-3 in fish oil has a potential effect on asthma. Thestudy aimed to determine the effect of illness perception on improvement symptoms with fish oil, measured byAsthma-Control-Test (ACT). This research was pre-post test design and purposive sampling, used 1 g daily fish oilthen followed-up after 4 weeks, conducted in March 2017–January 2018. The relationship between ACT value andillness perception using chi square test. The study sample consisted of 26 patients. The effectiveness of omega-3showed that there was significant difference of symptoms after the therapy. It can be concluded there was effect ofillness perception relationships related to symptoms. Most of the highest illness perception regarding symptoms andsymptoms related to asthma respondents experienced on the individual values, related to symptoms are fixed. Whilethe influence of illness perception on symptoms that got significant results was how long the asthma suffered, and theillness perception on causes of asthma that were significant to changes in asthma symptoms. Omega-3 fish oil waseffective in improving asthma symptoms, but the influence of illness perception must also be a concern.

ABSTRACT

High intake of omega-3 fatty acids has been associated with synaptic plasticity, neurogenesis and memory in several experimental models. To assess the efficacy of fish oil supplementation on oxidative stress markers in patients diagnosed with probable Alzheimer´s disease (AD) we conducted a double blind, randomized, placebo controlled clinical trial. AD patients who met the inclusive criteria were given fish oil (containing 0.45 g eicosapentaenoic acid and 1 g docosahexaenoic acid) or placebo daily for 12 months. Oxidative stress markers [lipoperoxides, nitric oxide catabolites levels, oxidized/reduced glutathione ratio, and membrane fluidity] and fatty acid profile in erythrocytes were assessed at enrollment, and 6 and 12 months after the start of the testing period. At the end of the trial, in patients who received fish oil, we detected a decrease in the omega 6/omega 3 ratio in erythrocyte membrane phospholipids. This change was parallel with decreases in plasma levels of lipoperoxides and nitric oxide catabolites. Conversely, the ratio of reduced to oxidized glutathione was significantly increased. In addition, membrane fluidity was increased significantly in plasma membrane samples. In conclusion fish oil administration has a beneficial effect in decreasing the levels of oxidative stress markers and improving the membrane fluidity in plasma(AU)


El alto consumo de ácidos grasos omega-3 se asocia con la plasticidad sináptica, neurogénesis y memoria en varios modelos experimentales. Para evaluar la eficacia de la suplementación con aceite de pescado en los marcadores de estrés oxidativo en pacientes con diagnóstico de la enfermedad de Alzheimer (EA) probable realizamos un ensayo clínico doble ciego, aleatorizado, controlado con placebo. A los pacientes con la EA que cumplían los criterios de inclusión se les administró aceite de pescado (que contenía 0,45 g de ácido eicosapentaenoico y 1 g de ácido docosahexaenoico) o placebo diariamente durante 12 meses. Los marcadores de estrés oxidativo plasmático [niveles de lipoperóxidos y catabolitos del óxido nítrico, cociente de glutatión reducido a glutatiónoxidado) y fluidez de la membrana] y el perfil de ácidos grasos en los eritrocitos se evaluaron al inicio, 6 meses y alos 12 meses. Al final del ensayo, en pacientes que recibieron aceite de pescado detectamos una disminución en el cociente de ácidos grasos omega 6/omega 3 en los fosfolípidos de la membrana eritrocitaria. Este cambio ocurrió en paralelo a la disminución de los niveles plasmáticos de lipoperóxidos y catabolitos del óxido nítrico. Por el contrario, el cociente de glutatión reducido a glutatión oxidado se incrementó significativamente. Además, la fluidez de la membrana aumentó significativamente en las muestras analizadas. En conclusión, la administración de aceite de pescado tiene un efecto beneficioso al disminuir los niveles de marcadores de estrés oxidativo plasmático y mejorar la fluidez de la membrana plasmática(AU)

ABSTRACT

Pollutants in the environment and exposure to them lead to medical ailments around the world. The present studyaimed to evaluate the protective effect of evening primrose oil and fish oil against fenitrothion pesticide-inducedtoxicity in rats. Male Wistar rats weighing 150-220 g, were randomly distributed into seven groups, the firstserved as control. The second group received fenitrothion, the third and fourth groups were supplemented withfish oil and evening primrose oil, respectively. Rats of group five were exposed to fish oil and fenitrothion. Thesixth group was exposed to evening primrose oil and fenitrothion. Rats of the seventh group were exposed to fishoil and evening primrose oil and fenitrothion. The results showed that the activities of serum alanineaminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), gamma-glutamyltransferase (GGT), and the levels of bilirubin (BIL), total protein (TP), albumin (ALB), glucose (GLU),cholesterol (CHOL), triglycerides (TG), low-density lipoprotein cholesterol (LDL-c), very low-densitylipoproteins cholesterol (VLDL-c), and superoxide dismutase (SOD) were noticeably increased in ratsadministered fenitrothion. However, the level of serum high-density lipoprotein cholesterol (HDL-c) andglutathione (GSH) were markedly decreased. It was found that fish oil and evening primrose oil decreased thephysiological unsettling influences initiated by fenitrothion. Moreover, the antioxidant properties of these oilssupported the bioactive roles of its defensive impacts on fenitrothion toxicity. Finally, the present findings suggestthat these oils may be utilized as preventive components against the toxicity of fenitrothion because of theirantioxidant properties.

ABSTRACT

Objective: To implement an evaluation system of functional food crude materials for their ranking to support the registration policy as required by legislation. Methods: Firstly, literature review and expert interview were used to determine the factors and their hierarchical relationships for the construction of the Functional Food Crude Materials Evaluation System (FUFMES). Secondly, the Analytic Hierarchy Process (AHP) method was used to calculate the weights of these factors. The specific method was used to construct a judgment matrix based on expert ratings, and R language was used to conduct consistency test and maximum characteristic root test was performed to obtain the weight of all levels of indicators. Thirdly, the extreme value was calculated for each factor; Fourthly, the index score and the ranking result were generated for each crude material; Lastly, the rankings obtained based on FUFMES were compared with that from the expert panel. Results: A total of 56 factors that were important for the evaluation were identified. These factors were organized in three levels. In particular, 6, 39 and 11 of them came from the first-level, second-level and third-level, respectively. The weights of these factors were determined using the AHP method. Nine crude materials were analyzed using FUFMES to generate the following index scores: Panax quinquefolium (0.49), Panax ginseng (0.48), Ginkgo biloba (0.21), Ganoderma lucidum spore powder (0.08), fish oil (0.06), Spirulina platensis (0.03), Coenzyme Q10 (0.02), melatonin (0.01), garlic oil (-0.03). Comparison of the rankings based on FUFMES index score and those by the experts showed high degree of consistency. Conclusion: FUFMES for integrated evaluation of the functional food crude materials were successfully established. FUFMES will become an invaluable tool for the ranking and evaluation of functional food crude materials catalogue and provide scientific basis and technical guarantee for the promotion of functional food crude materials registration.

Objectives: We studied the effects of fish oil and sake lees combined with a high fat diet in rats, and assessed serum lipids concentrations and anxiety behavior. Methods: Male Sprague-Dawley rats aged 5 weeks were fed a diet containing the Soybean oil (Control), Soybean oil + Sake lees (S), Fish oil (FO) or FO + Sake lees (FO+S) for 18days. The control diet contained a soybean oil component. It was measured the concentrations of serum lipids and anxiety behavior. Results: The concentration of serum cholesterol were significantly lower in the FO+S group than in the Control group. The concentration of serum triglyceride and free fatty acids were significantly lower in the FO group and the FO+S group than in the Control group. In anxiety behavior, the stay time in the zone of open arm were significantly higher in the S group than in the Control group. Conclusion: These results suggested that the combination of fish oil and sake lees in the diet improved the concentration of total cholesterol in the serum, which should assist in the prevention and improvement of dyslipidemia. In addition, intake of sake lees is expected to be effective in suppressing anxiety behavior.

ABSTRACT

Este trabalho teve como objetivo avaliar o efeito da substituição da farinha e do óleo de peixe pelo concentrado proteico de soja e óleo de soja, na sobrevivência, no crescimento e na composição corporal dos camarões (Litopenaeus vannamei) produzidos em sistema de bioflocos (BFT). Foram formuladas cinco dietas, isoproteicas e isoenergéticas, com diferentes níveis de substituição da farinha e do óleo de peixe por concentrado proteico de soja e óleo de soja, sendo os tratamentos designados como: 0% (sem substituição), 25%, 50%, 75% e 100%. As rações foram elaboradas para conter aproximadamente 35% de proteína e 8% de lipídios. O experimento foi conduzido durante 49 dias, com juvenis com peso inicial de 2,93±0,83g, em sistema de bioflocos (BFT). Não foram encontradas diferenças significativas entre os tratamentos até 75% de substituição para as variáveis de ganho de peso, conversão alimentar e sobrevivência. O tratamento de 100% de substituição apresentou menor taxa de crescimento específico. O presente resultado sugere que, nas dietas para camarões criados em sistema bioflocos, a farinha e o óleo de peixe possam ser substituídos em até 75% por concentrado proteico de soja e óleo de soja, sem prejudicar o desenvolvimento dos animais.(AU)


The objective of this work was to evaluate the effect of the substitution of fishmeal and fish oil with soy protein concentrate and soybean oil on survival, growth and body composition of shrimp (Litopenaeus vannamei) produced in biofloc system (BFT). Five diets were formulated to be isoproteic and isoenergetic with different levels of substitution of fishmeal and fish oil with soy protein concentrate and soybean oil. The treatments were named as: 0% (without substitution), 25%, 50%, 75% and 100%. The diets were formulated to contain approximately 35% protein and 8% lipids. The experiment was conducted for 49 days, with juveniles with initial weight of (2.93±0.83g) in a biofloc system (BFT). No significant differences were found between treatments up to 75% of substitution for the variables of weight gain, feed conversion ratio, and survival. The 100% substitution treatment showed a lower specific growth rate. The present study suggests fish meal and fish oil can be substituted in up to 75% for soy protein concentrate and soybean oil, without harming the development of the shrimps when reared in biofloc system.(AU)

Objetivou-se estudar o efeito do ômega 3 e da vitamina B12 no espermograma, na histomorfometria dos órgãos reprodutivos e na temperaturas do corpo com termografia infravermelha em ratos Wistar. Utilizaram-se 16 ratos, em quatro grupos (n=4), que receberam injeções diárias por 30 dias, sendo: grupo controle - solução salina; grupo ômega 3 - óleo de peixe 1g/kg; grupo B12 - vitamina B12 3µg; e grupo ômega 3 + B12 - óleo de peixe 1g/kg e vitamina B12 3µg. Imagens termográficas de áreas do corpo foram obtidas. No 30º dia, os ratos foram sacrificados e realizaram-se as análises de morfologia espermática e histomorfometria. Os dados foram submetidos à análise de variância e ao teste de Tukey a 5%. A temperatura da superfície do escroto foi superior no grupo B12 (P<0,05). Não houve diferenças entre grupos (P>0,05) para temperaturas do globo ocular. Houve correlação entre temperatura da superfície do escroto e porcentagem de gota citoplasmática distal (P=0,678). A elevação da temperatura do escroto resulta no aumento da porcentagem de gotas citoplasmáticas distais. A temperatura do globo ocular não sofre influência significativa do ômega 3 e da vitamina B12. O ômega 3 reduz o epitélio seminífero, e a vitamina B12 minimiza esse efeito.(AU)


The objective of this study was to study the effect of Omega 3 and vitamin B12 on spermogram, histomorphometry of reproductive organs and body temperature with infrared thermography in Wistar rats. Sixteen rats were used in four groups (n= 4) who received daily injections for 30 days. Control Group - saline solution; Group Omega 3 - fish oil 1g/kg; Group B12 - vitamin B12 3μg and Group Omega 3 + B12 - fish oil 1g/kg and vitamin B12 3μg. Thermographic images of body were obtained. On the 30th day the rats were sacrificed and analyzes of sperm morphology and histomorphometry were performed. Data were submitted to analysis of variance and Tukey's test at 5%. The surface temperature of the scrotum was higher in group B12 (P< 0.05). There were no differences between groups (P> 0.05) for eyeball temperatures. There was a correlation between scrotal temperature and distal cytoplasmic droplet (P= 0.678). Elevation of scrotum temperature results in an increase in the percentage of distal cytoplasmic droplets. The temperature of the eyeball is not significantly influenced by Omega 3 and vitamin B12. Omega 3 reduces the seminiferous epithelium and vitamin B12 minimizes this effect.(AU)

ABSTRACT

The aim of this study was to evaluate the n-3 fatty acid supplementation effects in breast cancer patients. Methods: it was a prospective longitudinal study performed in breast cancer patients supplemented with fish oil during 8 weeks in neoadjuvant AC-T regimen. Anthropometric, Bioelectrical impedance (BIA) ((Lean Body Mass (LBM), and Phase Angle (PA)), food consumption, nutritional impact symptoms (NIS), metabolic and inflammatory profile were evaluated for 2 months. Results: supplemented patients presented weight gain of 1.4 kg, representing 1.8% of body weight. LBM was 42.3 Kg and PA 6.1º. Nausea, diarrhea and anorexia were the main NIS with reduction in incidence with the supplementation, but xerostomia and heartburn had increase. There was significant drop in hemoglobin values to 12.3 mg/ dL, metabolic parameters and nutritional prognosis (CRP/albumin ratio) were not altered. Cytokines levels of TNF-α and interleukin-6 did not change, however interleukin-2 have shown significant increasing. Conclusion: Body weight gain was noticed in breast cancer patients, independent of food consumption. Fish oil supplementation during neoadjuvant chemotherapy was able to reduce NIS incidence, but not avoiding the decrease of hemoglobin levels, but it keeps the maintenance of proinflammatory parameters, except for increased the cytokine IL-2 (AU)


O objetivo foi avaliar os efeitos da suplementação de ácidos graxos n-3 em pacientes com câncer de mama. Metodologia: estudo prospectivo longitudinal realizado em pacientes suplementados com óleo de peixe durante 8 semanas em quimioterapia neoadjuvante com regime AC-T. Antropometria e impedância bioelétrica (massa corporal magra (massa magra) - ângulo de fase (AP)), consumo alimentar, sintomas de impacto nutricional (SIN) e perfil metabólico e inflamatório foram avaliados. Resultados:Os pacientes apresentaram ganho de peso de 1,4 kg, representando 1,8% do peso corporal. LBM foi 42,3 kg e AP 6,1º. Náusea, diarreia e anorexia foram o SIN com redução na incidência com a suplementação, mas a xerostomia e a azia aumentaram. Houve uma queda significativa nos valores de hemoglobina para 12,3 mg/dL, parâmetros metabólicos e prognóstico nutricional (relação PCR/albumina) não foram alterados. Os níveis de citocinas de TNF-α e interleucina-6 não mudaram, entretanto a interleucina-2 mostrou um aumento significativo. Conclusão: O ganho de peso corporal foi observado nas pacientes com câncer de mama, independente do consumo alimentar. Durante a quimioterapia neoadjuvante com suplementação de óleo de peixe houve redução da presença de SIN, diminuição dos níveis de hemoglobina e manutenção de parâmetros bioquímicos e inflamatórios, exceto pelo aumento da citocina IL-2 (AU)

ABSTRACT

Multiple organ dysfunction syndrome (MODS) patients or intestinal failure due to infection and other factors,intestinal dysfunction can not complete the absorption of nutrients and electrolytes.Progressive malnutrition in severe patients usually develops rapidly.Early parenteral nutrition can significantly improve the prognosis of MODS patients.PN is now widely used in all kinds of severe patients,but parenteral nutrition-associated liver disease (PNALD) caused by parenteral nutrition is prevalent in such patients.The emergence of PNALD increases the difficulty of curing patients' diseases.Many studies have found thatω-3fish oil fat emulsion in nutrient solution is an effective way to prevent and improve PNALD.

ABSTRACT

Objective To evaluate the blood clearance,tissue uptake and distribution as well as safety of high dose-40 mg intravenous fish oil/'medium chain triglycerides (FO/MCT:2 ∶ 8,wt/wt) lipid emulsions iu mice by measuriug the contents of triglycerides (TG) and free fatty acids (FFA) level in blood.Methods FO/MCT emulsions were radiolabeled with nondegradable [3H] cholesteryl ether to trace core particle metabolism in C57BL/6J mice following a bolus injection.For high dose,TG was 40 mg,100 times of low dose.Blood samples were obtained within 25 min to analyze TG and FFA concentrations;extracted organs were used to measure the tissue distribution of lipid emulsions.Results No animal in either group died,and no fat overload syndrome evidence was found after high dose injection.Compared with low dose,high dose injection increased the plasma TG and FFA concentrations rapidly;the emulsions were cleared significantly slower during 25 min after administration in mice;blood has a higher uptake ratio in organs,but heart has a lower one;there was no significant difference in liver and lung uptake ratio between the two groups.Conclusions High dose injection of FO/MCT is not fatal in mice.The animals only experience hypertriglyceridemia and high level of free fatty acids during a significant slower blood clearance period compared to low dose.Although blood clearance is slower,the blood uptake rate increases for further metabolism and clearance.Heart and lung functions are not affected as a lower cardiac and pulmonary uptake;blood uptakes more emulsions to further metabolize.FO/MCT may not induce fat overload syndrome in mice when administered with a super high dose.

ABSTRACT

Objective To explore the effect of fish oil fat emulsion as perioperative nutritional support on patients with liver cirrhosis and portal hypertension.Methods Randomized controlled clinical trial was performed between September 2011 and September 2017 in patients with liver cirrhosis and portal hypertension who underwent pericardial devascularization and splenectomy.Hypocaloric total parenteral nutritional support (TPN) started from the first day after the operation for 5 consecutive days.Patients were divided into experimental group and control group according to thetype of fat emulsion used.43 patients in experimental group were applied for fish oil fat emulsion injection (10% Omegaven) + medium long chain structure fat emulsion (20%STG) and 42 patients in control group were applied for medium long chain structure fat emulsion (20%STG).Liver function (total bilirubin and alanine aminotransferase),nutrition index (serum albumin and prealbumin),inflammatory mediators (TNF-α,IL-6 and IL-10) were measured before and after the operation,and the clinical outcomes were observed.Results There was no statistically significant difference in liver function and nutritional indices between the experimental group and the control group (P>0.05).The inflammatory mediators like TNF-u,IL-6 and IL-10 on the first day after surgery were significantly higher than those before surgery in both groups [experiment group:(225.54±54.78) vs.(61.49±16.47),(74.94±6.36) vs.(39.84±2.77),(77.53±11.4) vs.(46.05±6.13) ng/L;control group:(229.26±62.15) vs.(63.48±13.76),(77.23±7.83)vs.(40.64±3.34),(73.89±7.97)vs.(44.88±5.72) ng/L;P< 0.01].With the progress of time,the proinflammatory factors like TNF-α and IL-6 decreased after the operation and the range of decrease was higher in experiment group than in control group [d4-d 1:(-56.88± 31.63) vs.(-35.96±20.02),(-13.52±5.20) vs.(-6.38±2.84) ng/L;d7-d1:(-150.67±42.58) vs.(-132.79±53.35),(-27.04±8.97) vs.(-20.85±6.38) ng/L;P< 0.05].The range of increase in anti-inflammatory media IL-10 was higher in experiment group than in the control group (d4-d1:(14.22±13.08) vs.(5.64±3.58) ng/L;d7-d1:(17.78±5.58) vs.(-37.96±11.43) ng/L;P<0.05).The incidence of grade Ⅲ complications and total complications (4.7% vs.21.4%,23.3% vs.45.2%) and hospitalization time [(10.12 ±1.48) vs.(12.33±2.04) d] in the experimental group were significantly lower than those in the control group (P<0.05).Conclusions In patients with liver cirrhosis and portal hypertension,perioperative nutritional support of fish oil fat emulsion can reduce systemic inflammatory response and operative complications and promote rapid recovery through its two-way regulation of inflammatory mediators.

ABSTRACT

PURPOSE: Glucocorticoids (GCs) are implicated in secondary osteoporosis, and the resulting fractures cause significant morbidity. Polyunsaturated fatty acids (PUFAs) play a vital role in bone metabolism. However, few trials have studied the impact of omega-3 PUFA-containing oils against GC-induced osteoporosis. Therefore, the present study was undertaken to determine whether supplementation with omega-3 PUFA-containing dietary oils such as fish oil, flaxseed oil or soybean oil can impede the development of GC-induced osteoporosis. METHODS: The fatty acids (FAs) content of oils was determined using gas chromatography. Male rats were subdivided into 5 groups (8 rats each): normal control (balanced diet), prednisolone control (10 mg/kg prednisolone daily), soybean oil (prednisolone 10 mg/kg + soybean oil 7% w/w), flaxseed oil (prednisolone 10 mg/kg + flaxseed oil 7% w/w), and fish oil (from cod liver; prednisolone 10 mg/kg + fish oil 7% w/w). RESULTS: The study data exhibited a significant depletion in bone mineral density (BMD) and femur mass in the prednisolone control compared to the normal control, accompanied with a marked decrease in the levels of plasma calcium and 1,25-(OH)₂-vitamin D₃, and elevated levels of C-terminal telopeptide (CTX), tumor necrosis factor-alpha (TNF-α) and malondialdehyde (MDA). Supplementation with fish oil, soybean oil or flaxseed oil helped to improve plasma calcium levels, and suppress oxidative stress and inflammatory markers. Additionally, bone resorption was suppressed as reflected by the decreased CTX levels. However, fish oil was more effective than the other two oils with a significant improvement in BMD and normal histological results compared to the normal control. CONCLUSION: This study demonstrated that supplementation with dietary oils containing omega-3 PUFAs such as fish oil, soybean oil or flaxseed oil can play a role in the prevention of bone loss and in the regulation of bone metabolism, especially fish oil which demonstrated a greater level of protection against GC-induced osteoporosis.

Objective@#To investigate the application value of ω-3 fish oil fat emulsion in the parenteral nutritional support treatment following radical gastrectomy for gastric cancer.@*Methods@#The retrospective cohort study was conducted. The clinical data of 60 patients who underwent radical gastrectomy for gastric cancer in Nanjing Medical University Affiliated Wuxi Second Hospital between January 2018 and December 2018 were collected. There were 37 males and 23 females, aged from 28 to 78 years, with an average age of 64 years. Thirty patients who received parenteral nutrition containing 100 mL of ω-3 fish oil fat emulsion after radical gastrectomy and 30 patients who received parenteral nutrition containing routine fat emulsion after radical gastrectomy were allocated into experimental group and control group, respectively. Observation indicators: (1) nutritional indicators in the perioperative period; (2) inflammatory indicators in the perioperative period; (3) immune indicators in the perioperative period; (4) postoperative complications. Measurement data with normal distribution were represented as Mean±SD, and comparison between groups was evaluated using the independent-sample t test. Count data were described as absolute numbers and percentages, and comparison between groups was analyzed using the chi-square test. Repeated measurement data were analyzed by the repeated measures ANOVA.@*Results@#(1) Nutritional indicators in the perioperative period: the levels of total protein, albumin, prealbumin, and transferrin from preoperative day 1 to preoperative day 6 were respectively changed from (60.2±3.0)g/L to (57.2±3.1)g/L, from (35.3±3.1)g/L to (37.0±1.8)g/L, from (186±24)mg/L to (172±17)mg/L, from (3.0±0.7)g/L to (2.4±0.4)g/L in the experimental group and from (60.6±2.4)g/L to (55.7±4.2)g/L, from (35.0±3.8)g/L to (36.0±3.8)g/L, from (184±18)mg/L to (173±25)mg/L, from (3.1±0.6)g/L to (2.2±0.8)g/L in the control group, with no significant difference in the changing trends between the two groups (F=0.79, 2.14, 0.03, 0.36, P>0.05). (2) Inflammatory indicators in the perioperative period: the levels of white blood cells, C-reactive protein, interleukin 6, and tumor necrosis factor-α from preoperative day 1 to preoperative day 6 were respectively from (7.2±1.1)×109/L to (10.2±0.9)×109/L, from (7.2±2.3)mg/L to (25.5±6.3)mg/L, from (16±3)ng/L to (24±4)ng/L, from (17±4)ng/L to (22±5)ng/L in the experimental group and from (7.4±0.8)×109/L to (13.0±1.3)×109/L, from (6.9±2.4)mg/L to (41.6±18.9)mg/L, from (17±4)ng/L to (45±8)ng/L, from (16±4)ng/L to (43±7)ng/L in the control group, respectively, with significant differences in the changing trends between the two groups (F=63.05, 51.65, 127.82, 104.91, P<0.05). (3) Immune indicators in the perioperative period: the levels of immunoglobulin A, immunoglobulin G, immunoglobulin M, CD4+, CD8+, and ratio of CD4+ /CD8+ from preoperative day 1 to preoperative day 6 were respectively from (1.5±0.4)g/L to (2.8±0.5)g/L, from (11.1±1.7)g/L to (14.0±1.2)g/L, from (0.77±0.28)g/L to (1.61±0.31)g/L, from 42%±6% to 46%±5%, from 23%±4% to 24%±3%, from 1.82±0.42 to 2.11±0.24 in the experimental group and from (1.4±0.4)g/L to (2.3±0.6)g/L, from (10.7±1.8)g/L to (11.9±1.4)g/L, from (0.69±0.23)g/L to (1.19±0.33)g/L, from 40%±5% to 39%±4%, from 24%±3% to 23%±3%, from 1.75±0.34 to 1.81±0.35 in the control group, respectively, showing significant differences in the changing trends of the levels of immunoglobulin A, immunoglobulin G, immunoglobulin M, CD4+, and ratio of CD4+ /CD8+ between the two groups (F=18.39, 15.20, 38.42, 9.55, 5.50, P<0.05), showing no significant difference in the changing trend of the levels of CD8+ between the two groups (F=0.89, P>0.05). (4) Postoperative complications: 5 patients had postoperative complications, with a incidence rate of 16.7%(5/30), including 1 of abdominal infection, 1 of incisional infection, and 3 of pulmonary infection, and all the 5 patients were cured after symptomatic treatment. Nine patients had postoperative complications, with a incidence rate of 30.0%(9/30), including 2 of abdominal infection, 2 of incisional infection, and 5 of pulmonary infection, and all the 9 patients were cured after symptomatic treatment. There was no significant difference in the incidence of postoperative complications between the two groups (χ2=1.491, P>0.05).@*Conclusion@#For patients who receive gastric cancer surgery, ω-3 fish oil fat emulsion can reduce the inflammatory response, improve their immune function and not increase postoperative complications.

El síndrome metabólico (SM) constituye una constelación de factores interrelacionados que elevan el riesgo de padecer enfermedad cardiovascular y diabetes mellitus tipo 2. La composición de la dieta es crucial en la modificación de estos factores. Numerosos trabajos focalizaron su atención en fuentes de ácidos grasos n-3 de origen marino (animal) en el manejo de estos trastornos, pero más escasos y contemporáneos son aquellos de fuentes n-3 de origen vegetal.
